Expression of serum miR-300 in cholangiocarcinoma patients and clinical significance

Abstract
Objective To detect the expression of serum miR-300 in cholangiocarcinoma patients and discuss its clinical significance. Methods Serum samples of 107 cholangiocarcinoma patients and 107 normal control patients were collected. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to detect the expression of serum miR-300. Diagnostic value of serum miR-300 in cholangiocarcinoma was analyzed by the receiver operating characteristic (ROC) curve. Associations between serum miR-300 expression and clinicopathologic features of cholangiocarcinoma patients were analyzed by χ² test. Association of serum miR-300 expression and 5-years overall survival of cholangiocarcinoma patients were analyzed by Kaplan-Meier methods. Results The expression of serum miR-300 in the cholangiocarcinoma patients was statistically higher than that of the normal controls ( < 0.05), moreover, the expression of serum miR-300 was significantly down-regulated after surgery. The expression of serum miR-300 in the cholangiocarcinoma patients was significantly associated with clinical stage ( = 0.010), but had no significant association with age, gender or differentiation status of cholangiocarcinoma. Cholangiocarcinoma patients with high serum miR-300 expression had a significantly lower 5-year overall survival rate compared to those with low serum miR-300 expression. Conclusions The expression of serum miR-300 is significantly up-regulated in cholangiocarcinoma, and serum miR-300 is a promising diagnostic and prognostic biomarker in cholangiocarcinoma.关键词
