Abstract

Objective:To screen out key molecular for hepatocellular carcinoma(HCC) STAT3 signaling pathways and to study the expression quantity change of caspase3,muc1 and cyclinD1 in HCC after the STAT3 downregulated.Method:From May 2014 to May 2016, 40 liver cancerpatients of our hospital for treatment were as the research objects,and liver cancer tissue and normal adjacenttissue to carcinoma taken respectively from these people were as the research material.The techniques of immunofluorescence and Western blot were used to analyze the expression of STAT and P-STAT3 proteinin tumor tissues and cells.According to the GENEBANK of coding sequence of STAT3,and screening out target sequence,the same target sequences were taken as potential siRNA targeted point.Multiple target sequences of siRNA were designed after the BLAST comparison,modifiedwith fluorescein markers,then transfected in cell lines,to screen out transfection most efficient siRNA sequences.Using RT-PCR technique analyzing the mRNA level on expression quantity of caspase3,muc1,cyclinD1,P21 and MMP2 in human liver cell lines.Western blot was used to analyze the expression of the STAT and P-STAT3 protein after transfection cell lines.Result:We found that STAT3 and P-STAT3 protein expression content in cancer of the liver tissue was higher than that in normal tissue adjacent to carcinoma by Western blot to test the expression of STAT3 and P-STAT3 protein respectively in HCC tissue and normal tissue adjacent to carcinoma.Downregulation STAT3 could inhibit the Hep-G2 proliferation in liver cancer cell and invasion abilityin vitro,promoted apoptosis, and made STAT3 gene silence.STAT3 signal molecules on the signaling pathway also significantly lower expression at different time points.Conclusion:We find that STAT3 downregulation could inhibit the biological behaviour influence in the liver cancer cell proliferation and colonization.We speculate that STAT3 might be the new molecular targets of liver cancer in the treatment of HCC.
