摘要

Abstract

Objective To establish a method for microbial limit test on tigecycline. Methods The sample (5g) was dispersed uniformly to the nutrient broth medium (100mL),which was centrifuged for 3 minutes at 500r/min.The supernatant was diluted with the ratio of 1:20 as the test solution,and the filter membranes were incubated with the test solution (1mL) respectively,which was rinsed by the sterile solution(600mL) containing 0.3%tween-80 and 0.1%peptone,with the aim of eliminating the interference of tigecycline's bacteriostatic activity,to verify the method of Tigecycline's microbial limit test. Results With the method above,the recoveries of all the five validated bacteria were over 70%. Conclusion The membrane filtration method,which is that the membrane-filter is rinsed by the sterile solution (600mL) containing 0.3% tween-80 and 0.1% peptone,and it can be applied for the test of Tigecycline’s microbial limit.
