Abstract

Objective To investigate the prevalence of swine HEV infection in rural areas of Dali City in Yunnan province, where raw pork is traditionally consumed. Methods HEV ORF2 was detected in swine stool samples ( n =41 ) by reverse transcription⁃nested PCR ( RT⁃nested PCR) . The PCR products were detected by electrophoresis on agarose gel and purified, and then cloned into pMD18⁃T vector and sequenced. The homology and phylogenetic analysis were performed by DNAStar and MEGA4. 0 software, respectively. Results The target gene (348 bp) of HEV was amplified by RT⁃nested PCR. Phylogenetic analysis showed that these isolated viruses were swine HEV genotype 4 and h subtype. In this study, 23 samples were found to have swine HEV, and the positive rate was 56. 10 % ( 23/41 ) . Conclusion Swine HEV infection was highly prevalent in Dali, which indicated a high risk of zoonotic transmission of HEV from swine to humans. Therefore, strategies should be performed to prevent against HEV infection.
