桑蚕茧丝素蛋白肽对大鼠离体过氧化肝组织的修复及对小鼠过氧化肝损伤的预防作用观察
Effects of silk protein polypeptides of silkworm cocoon on repairing isolated oxidative liver tissues in rats and its preventive effect on oxidative liver injury in mice

Abstract
Objective To observe the effect of silk protein polypeptides of silkworm cocoon (SFP)on repairing oxida-tive liver tissues in rats and its preventive effect on oxidative liver injury in mice.Methods ①Anti-oxidation effect of SFP on the isolated rat liver:three Sprague-Dawley rats were sacrificed and prepared the liver microsomes,after adding ascorbic acid and ferrous ion solution to induce oxidation,they were divided into 5 groups,0.2 mL SPF was added to the experi-mental groups 1,2 and 3 with concentrations of 10,15 and 30 ug/mL,respectively,the equal amount of 10 g/mL antioxi-dant BHT and distilled water were added to the positive control group and blank control group,respectively.We measured the content of MDA and calculated the inhibition rate of oxidation after incubation in 37 ℃ water for 2 h.② Intervention effect of SFP on liver injury induced by oxidation in mice:Fifty healthy BALB/C mice were randomly divided into groups A,B,C,D and E.Mice in the groups A,B and C were injected with 37.5,75 and 112.5 mg/kg SFP once a day;mice in the group D were injected with normal saline once a day and in group E were fed normally.After 33-day administration, mice were starved overnight and injected as described above,after 0.5-1 h mice in the groups A,B,C and D were given brominated oil to produce the liver injury models by oxidation.After 24 h,mice were sacrificed and we took the liver tis-sues to measure MDA,SOD and GSH-Px.In the experiment process,we recorded the body weight of mice every day and observed the spirit,diet and activity of mice.Results The content of MDA in the liver tissues of the experimental groups 1,2,3 and positive control group was lower than that of the blank control group,and the MDA content in the experimental groups 1,2 and 3 was decreased gradually (all P <0.05).The inhibition rates of lipid peroxidation in the experimental groups 1,2 and 3 were increased gradually and they were all lower than that of the positive control group (all P <0.05). The contents of MDA in serum and liver tissues of groups A,B and C were lower than those in the groups D and E,and the activity of SOD and GSH-Px in serum and liver tissues was higher than that of groups D and E.The contents of MDA in the groups A,B and C were reduced,while the activity of SOD and GSH-Px was increased gradually (all P <0.05).During the experiment,the mice did not appear weight loss,mental malaise or even death.Conclusions SFP could repair the oxi-dative liver injury of rats in vitro and prevent liver injury in mice,which is in a dose-dependent manner.The effect of SFP on liver injury may be related to reducing the formation of peroxidation products and improving antioxidant enzyme activity.关键词
