miR-101对上皮性卵巢癌SKOV3细胞抗失巢凋亡的作用研究
Study on the role of miR-101 on anoikis resistance of epithelial ovarian cancer SKOV3 cells

Abstract
Objective:To investigate the role of miR-101 on anoikis resistance of epi-thelial ovarian cancer SKOV3 cells and the possible mechanisms of this process. Methods:We established an anoikis pattern to imitated the growth status of cancer cells metastasis. miR-101 mimics and miR-NC were transfected into epithelial ovarian cancer SKOV3 cells by using li-posomes Lipofectamine 2000 , and then SKOV3 cells were randomly divided into the control group,the miR-blank plasmid group ( miR-NC group) and the miR-101 plasmid group. The ex-pression level of miR-101 and the rate of apoptosis in the three groups were detected by RT-PCR and Flow Cytometry after transfection. The ability of cell invasion and migration was meas-ured by Transwell invasion assay and Wound-healing assay,respectively. The messenger RNA and protein expression level of c-fos were respectively measured by RT-PCR and Western blot. Results:The anoikis pattern was established successfully,and the SKOV3 cells were suspended and consolidated into multicelluar spheroids. The expression level of miR-101 was increased af-ter miR-10 mimics was transfected into SKOV3 cells, and the cells anoikis increased, the a-mounts of invasive cells and the migration distance of cells was reduced. In addition,the mes-senger RNA and protein expression level of c-fos was decreaseed. Conclusions:miR-101 over-expression could increase the anoikis of SKOV3 cells,and induce its ability of invasion and mi-gration. The down-regulation of c-fos expression may be the mechanism of its action.关键词
