Rsf-1/HBXAP基因在宫颈癌中的表达及临床病理意义
Research of Rsf-1/HBXAP expression and clinical pathological characteristics in cervical cancer

Abstract
Objective:To investigate the expression and significance of remodeling spacing factor (Rsf-1/HBXAP) in cervical cancer,and to analyze the gene detection methods and theoretical basis for the cervical cancer targeted treatment. Methods:The immunohisto-chemical staining was used to detect Rsf-1/HBXAP expression in 17 cases of normal control,35 cases with cervical intraepithelial neoplasm ( CIN) and 182 cases with cervical cancer. RNA were extracted from HeLa,Siha and 293T by the routine phenol-chloroform method,which were used for PCR amplification. Besides, the expression levels of Rsf-1/HBXAP in the cell lines were also detected by Western blot. Finally,Kaplan-Meier survival analysis was used to evaluate the difference of overall survival between the high Rsf-1/HBXAP expression and low Rsf-1/HBXAP expression. Results:The positive rates of Rsf-1/HBXAP were 5. 88% in normal cervi-cal tissues,37. 14% in CIN lesions,79. 12% in carcinoma respectively,which was significantly different ( P<0 . 05 ) . The positive expression rate of Rsf-1/HBXAP in cervical cancer tissues was correlated with the degree of tumor differentiation and clinical staging ( P<0 . 05 ) , which was no related to age and size ( P>0 . 05 ) . Rsf-1/HBXAP mRNA expression and protein expres-sion in HeLa and Siha were higher than 293T. Kaplan-Meier survival analysis revealed that RSF-1 overexpression is significantly related to poor clinical outcome for the women suffering cervical carcinoma. Meanwhile, Fisher′s exact test and X2 test also confirmed these findings. Conclusion:Rsf-1/HBXAP closely associated with precancerous lesions and cervical cancer, and might provide new information regarding the treatment strategies for cervical cancer combi-nation therapy.关键词
