利拉鲁肽对大鼠2型糖尿病合并非酒精性脂肪肝内质网应激的影响研究
Effects of Liraglutide on Endoplasmic Reticulum Stress in Mice with T2DM and Fatty Livers

Abstract
Objective To investigate the effects of liraglutide on endoplasmic reticulum stress ( ERS ) in mouse with T2DM and fatty livers. Methods SD mice of 8 weeks were subdivided into four groups:the first group was given regular diet ( NC group, n=10 ) , the second group received high fat and sugar diet ( MC group, n=9 ) , the third group was given high fat and sugar diet and low dose of liraglutide ( GLP-L, n=10 ) and the forth group was given high fat and sugar diet and high dose of liraglutide ( GLP-L, n=8 ) . The rats in GLP-L group and GLP-H group were given 0. 4 mg/kg and 0. 8 mg/kg of liraglutide by subcutaneous injection from the 9th week. The mRNA expression of genes ( SREBP1, EDEMI and apoB100 ) were measured by quantitative real-time PCR. Results Fat metabolism ( FBG ) and fasting plasma insulin ( FINS ) in MC group were significantly higher than those in NC, GLP-L and GLP-H group, and Plasma total cholesterol ( TC ) , triglyceride ( TG ) , high-density lipoprotein ( HDL-C ) , free fatty acids ( FFAs ) and HOMA-IR in MC group were also significantly higher than those in MC , GLP-L and GLP-H group ( P ﹤ 0. 05 ) . However, Low density lipoprotein ( HDL-C ) in MC group was lower than that in NC, GLP-L and GLP-H group ( P ﹤ 0. 05 ) . FBG and FINS in GLP-H group were significantly lower than those in GLP-L ( P ﹤ 0. 05 );compared with NC group, the expression of SREBP1, EDEMI and apoB100 mRNA were higher significantly up-regulated ( P ﹤ 0. 05 ) , but the above indicators in GLP-L and GLP-H group were obviously lower than those in MC group ( P ﹤ 0. 05 ) , additionally, the expression of SREBP1 in GLP-H group were significantly lower than that in GLP-L ( P ﹤ 0. 05 ) . Conclusion ERS plays a central role in development of liver steatosis, but liraglutide may reduce the activation of ERS in T2DM mouse then slow the process of NAFLD incidence.关键词
