Abstract
Objective To investigate the differences of Th 17 cell-mediated immunological responses between children with airway inflammation diseases and healthy controls . Metho ds 41 asthmatic children ( AS) , 40 children with allergic rhinitis ( AR) , 52 children suffering with asthma combined with allergic rhinitis ( ASR) and 45 sex matched healthy controls ( HC) were enrolled in this study .The percentages of circulating Th1 cells and Th17 cells were determined by flow cytometry .The level of IL-17A, IL-17E and IL-17 F in plasma or in mite activated PBMCs supernant was measured by using enzyme-linked immu-nosorbent assay .The correlationship between Th 17 cells and related cytokines , fractional exhaled nitric oxide levels ( FeNO) and peripheral blood eosinophils counts ( EOS) was analyzed .Results The per-centage of Th1 cell population in ASR group , AR group and AS group was 13.2%±5.3 %, 12.2%± 6.7%and 12.1%±4.1% respectively , which were significantly lower than in HC group ( 17.0% ± 5.1%) .The proportion of Th 17 cells in peripheral blood was highest in ASR group (1.8%±0.7%) , fol-lowed by AS group (1.2%±0.5%) and AR group (1.1%±0.6%), which were significantly higher than in HC group (0.5%±0.4%, P<0.01).The levels of IL-17A and IL-17E of three patient groups were significantly higher than in HC group , the inter-group differences were statistically significant ( P<0.01).IL-17E was produced by PBMCs in AS group, AR group and ASR group after stimulation with mite in vitro.The IL-17E levels of patient groups were positively correlated with elevated FeNO levels and in -creased EOS in peripheral blood (P<0.05, respectively).Only the Th17 cells in ASR group was positive-ly correlated to elevated FeNO level and increased EOS in peripheral blood ( P<0.05 ) .Conclusion Th17 cell and its effector cytokine , IL-17E, have played an important role in the development of allergic airway inflammation .关键词
