摘要

Abstract

Objective To explore the effect of lipoxin A4 ( LXA4 ) on the proliferation of hu-man hepatocellular carcinoma ( HCC) under hypoxic condition. Methods Ectopic HCC mice mo-del was established by injection H22 cells subcutaneously. Immunohistochemistry assay was applied to detect the protein expression of Ki-67 and hypoxia-inducible factor-1 α ( HIF-1α) . HepG2 cells were cultured under 1% O2 to establish a hypoxic condition. 3-( 4,5-dimethyl-2-thiazolyl)-2,5-di-phenyl-2-H-tetrazolium bromide ( MTT) assay was used to measure cell proliferation rate. Results In 7 to 15 days after H22 cell transplantation,the subcutaneous tumor in mice could be observed. At all the time points, the tumor volume of LXA4 mice was significantly reduced (P<0. 05). At the same time, LXA4 significantly decreased the expression of Ki-67 and HIF-1αinside tumor tissue. In vitro study showed that 200 nmol/L LXA4 obviously inhibited HepG2 proliferation under 1% O2 . Conclusion LXA4 could inhibit the HCC growth under hypoxia.
