蕨麻多糖对缺氧损伤人脐静脉内皮细胞的保护作用及机制
Protective effect of Potentilla anserine L.polysaccharide on human umbilical vein endothelial cells after hypoxia injury

Abstract
Objective To investigate the protective effect and mechanism of Potentilla anserine L.polysaccharide on human umbilical vein endothelial cells injured by hypoxia.Methods The cultured human umbilical vein endothelial cells (EA.hy926)were randomly divided into the normoxic control group,hypoxia model group,Fufangdanshen control group (with 0.1 mg/mL Fufangdanshen tablet extract)and high-,medium-and low-concentration Potentilla anserine L.polysac-charide groups (with 0.1,0.05,and 0.025 mg/mL Potentilla anserine L.polysaccharide,respectively).In addition to the normoxic control group,the hypoxia injury models were constructed in the other five groups.The cells were cultured for 2 h.The metabolic activity and the survive rate of the cells were determined with MTT assay and Trypan-blue staining.The morphological changes of the cells were observed under light microscope.The levels of lactate dehydrogenase (LDH)and nitric oxide (NO)in the culture medium were detected by colorimetric method as well as the levels of superoxide dismutase (SOD)and the content of malondialdehyde (MDA)in cells.The levels of the inducible NO synthase (iNOS)and the en-dothelial NO synthase (eNOS)and the endothelin 1(ET-1)in culture medium were measured by ELISA.The expression levels of iNOS mRNA and eNOS mRNA was detected by RT-PCR.Results Comparing with hypoxia model group,the morphology of the cells were improved significantly,the cell viability and survival rate increased,cell LDH leakage de-creased,iNOS and ET-1 levels decreased,eNOS and NO levels increased,SOD levels increased,MDA levels decreased, cell eNOS mRNA expression increased,and iNOS mRNA expression decreased in the Fufangdanshen control group and each Potentilla anserine L.polysaccharide group (all P <0.05).Conclusion The Potentilla anserine L.polysaccharide could have protective effect on the endothelial cells during hypoxia by improving anti-oxide activity of the cells,and the effect might be related to its regulation of dynamic equilibrium between ET-1 and NO.关键词
