Abstract

In this paper,we investigated the influence of short-term shading on the biosynthesis of purine alkaloids,amino acids and catechins.We analyzed the content of three major metabolites (the metabolisms of purine alkaloids,amino acids and polyphenols) and the expression of key genes in biosynthesis pathways in tea (Camellia sinensis) by HPLC (high performance liquid chromatography) and fluorescence quantitative PCR.Our data revealed that short-term shading slightly down-regulated the total catechins and restrained the biosynthesis of caffeine.Moreover,short-term shading had notable effects on amino acids biosynthesis by increasing 58.5%.The content of theanine had about sixfold increase compared with the control group,and it was remarkably correlated with the expression of some genes,such as TCS1,PAL,F3'H and ANR2 according to the differential expression of these genes.The results showed that shading has certain relation with the biosynthesis of purine alkaloids,amino acids and polyphenols in rooted tea cuttings.
