Influence of co-culturing human umbilical vein endothelial cells with T24 bladder cancer cells on the invasion, migration and proliferation of T24 cells

Abstract
Objective:To assess the effects of co-culturing human umbilical vein endothelial cells (HUVEC)with T24 bladder cancer cells on the invasion,migration and proliferation of T24 cells.Methods:We divided the study into two groups:a T24 bladder cancer cell group (T24 group) and the HUVEC-T24 bladder cancer cells trans-well co-culture group (co-culture group).The cell invasion,migration ability,and proliferation were compared using trans-well cell invasion Matrigel assay,transwell migration assay and CCK-8 proliferation assay.Results:After 24 hours,the number of invasive and migrated cells in the co-culture group were respectively 93.22±27.35 and 183.62±62.07,which were significantly higher than those in the T24 group (52.04±22.54 and 66.80±23.36) (P <0.01).The CCK-8 assay showed that the OD values on day 3,4 and 5were respectively (1.128±0.125),(0.866±0.068) and (1.536±0.098) in the co-culture group,and were (1.103±0.153),(1.910±0.139) and (1.550±0.177) in the T24 group.The cell proliferation rate was markedly higher in the co-culture group than the T24 group (P <0.01).Conclusion:Co-culture of HUVEC with T24 bladder cancer cells promotes the invasion,migration and proliferation of T24 cells.关键词
