脱氧雪腐镰刀菌烯醇的高灵敏化学发光磁酶免疫法检测
Determination of deoxynivalenol by a sensitive method of magnetic en-zyme chemiluminescence immunoassay

Abstract
Aim: To propose a magnetic enzyme chemiluminescence immunoassay ( MECLIA ) for rapid and sensitive detection of deoxynivalenol (DON).Methods: Fe3O4@SiO2@Au particles and the conjugation of DON to ovalbumin (OVA-DON) were prepared.Then,the DON-OVA was coated on the surface of Fe3O4@SiO2@Au by using Fe3O4@SiO2@Au as immobile phase.After that, MECLIA based on the luminol-H2O2-HRP-phenols system for detection of DON was established.The accuracy and sensitivity of the established method were evaluated .Results:The standard curve equation was Y=-521951 X+2883979 ( X was the logarithm of the concentration of DON , Y was the illumination value of the standard concentration, R2 =0.9986), the linear range was 0.10~20.00 μg/L, the limit of detection was 31 ng/L, the relative standard deviations were less than 3.5% and 9.6% for intra-and inter-assay, respectively, and the recoveries ranged from 95.0%to 108.0%.The proposed method was satisfactorily applied to determine DON in wheat sample and the recoveries ranged from 92.0%to 106.0%.Conclusion:The developed MECLIA is sensitive and simple with good preci-sion for DON residue analysis in wheat samples.关键词
