Abstract
Aim: To screen the activity of 2-methoxyestradiol (2-ME) and synthetic derivatives by rat lung cell mem-brane chromatography .Methods:The lung cell membrane stationary phase ( CMSP ) was packed into a stainless steel col-umn(10 mm ×2 mm), and the HPLC conditions were as follows:a mobile phase consisted of 10 mmol/L phosphate buffer (pH 7.4) at a flow rate of 0.2 mL/min, and the column temperature was 37 ℃.Salbutamol was used as positive control in the screening .The capacity factor k′as chromatographic parameters was used to describe the affinity of 2-ME and its synthetic derivatives .Results:2-ME,NO.1 synthetic derivative and NO .2 synthetic derivative had chromatographic reten-tion on lung CMSP column and k′of NO.2 was greater than NO.1 and 2-ME.The k′of 2-ME with rat lung CMSP was higher than those with pure silica gel column ,rat cardiac muscle CMSP and liver CMSP .Conclusion:2-ME has the proper-ty of specific affinity on the membrane of the rat lung cell membrane.关键词
