摘要

Abstract

Objective To explore protective effects of hydroxysafflor yellow A (HSYA) in neonatal rat primary cadiocyte anoxia-reoxygenation (A/R) injury were relevant to inhibiting the activation of Mst1. Methods The neonatal rat cadiocyte was isolated. Incubation of 48 h, cell attachment, and then beginning group experiments. Normal control group (Con group ): the neonatal rat cadiocyte were cultured under normal conditions for 36 h; Anoxia-reoxygenation group (A/R group): after incubation with normal 24 h, hypoxia 10 h reoxygenation 2 h; Hydroxy safflor yellow A treatment group (H + A/R group): after incubation with 24 h, hypoxia 10h was added with HSYA at the same time, and then reoxygenation (2 h); Mst1siRNA transfection group: after transfection of small interfering 6h, for normal medium, incubated with 18h, hypoxia 10h reoxygenation 2h. Cadiocyte cell culture supernatant was collected,to detect LDH.The level of reactive oxygen species and apoptosis rate were detected by flow cytometry. Western blot detected Mst1, Cleaved caspase-3, Caspase-3 changes in protein expression. Results Compared with the control group,the increase of ROS, the apoptosis rate increased, the expression of activated Mst1 increased, the expression of cleaved caspase-3 protein increased in A/R group, the difference was statistically significant;Compared with A/R group, the level of ROS decreased, the apoptosis rate decreased, the expression of p-Mst1 protein decreased, and the expression of cleaved caspase-3 protein decreased in the A/R+H group, the difference was statistically significant.Compared with A/R group, there was no significant change of ROS, the apoptosis rate decreased, the total Mst1 and p-Mst1 protein expression decreased, cleaved caspase-3 decreased in the Mst1siRNA group, the difference was statistically significant; Compared with the A/R+H group, the level of ROS was higher, and the apoptosis rate of myocardial cells was increased in the Mst1siRNA group, and the difference was statistically significant. Conclusion HSYA can reduce the emergency of the oxidant stress and decrease the apoptosis of the cells,and the mechanism may dependent on the effect to inhibit the activation of Mst1, but there are other channels.
