RNAi慢病毒表达对小鼠海马CA1神经元电生理功能的影响
The effect of expression of RNA interfering lentivirus in the hippocampus of mice on the electrophysiological functions of hippocampal CA1 pyramidal neurons

Abstract
U6 promoter and enhanced green fluorescent protein (pU6-EGFP)-containing RNA interfering lentivirus was injected into the hippocampal CA1 region for 2-3 weeks by the stereotaxic,then we used patch clamp and field potentials to study the changes in the electrophysiological property,synaptic transmission and synaptic plasticity of hippocampal CA1 pyramidal neurons.The result showed that:pU6 EGFP lentivirus injection did not affect the basal synaptic transmission,the synaptic plasticity,the membrane potential and the discharge characteristics of CA1 pyramidal neurons (resting membrane potential,threshold of action potential,the current to evoke action potential) in the Schaffer collateral-CA1 pathway.The data demonstrated that pU6-EGFP lentivirus injection had no effect on the electrophysiological functions of hippocampal slices,so it is a feasible strategy to apply the pU6-EGFP lentivirus in the functional study of central nervous system.关键词
