Abstract
Objective To explore the relationship between blood stasis condition and cytokine/NF-κB signaling path-way in patients with ankylosing spondylitis and the intervention mechanism of Xinfeng capsule. Methods A total of 42 patients with ankylosing spondylitis were selected as observation group, and 40 healthy subjects were enrolled as control group. Observation group taked Xinfeng capsule, each 1. 5 grams, 3 times every day, continuous medication for 3 months. The blood immune and blood stasis cytokine were detected by ELISA, and NF-κB signaling pathway in-dicators gene and protein expression were measured by RT-PCR and Western-blot. Results Compared with control group, the content of PAI-2, TNF-α, TXA2, PGI2, GMP140, IL-1βand IL-6 in observation group significantly in-creased pre-treatment(P<0. 05), and IL-4, IL-10 significantly decreased(P<0. 05). The content of PAI-2, TNF-α, TXA2, PGI2, GMP140, IL-1β and IL-6 significantly decreased after the observation group were treated with the Xinfeng capsule treatment for 3 months(P<0. 05), and IL-4, IL-10 significantly increased(P<0. 05). The gene expression of IκBα, IKKβ, p65, p50 mRNA and the protein expression of p65, p50 in the observation group before treatment showed higher than control group(P<0. 05), and those showed lower after treated with the Xinfeng capsule treatment for 3 months(P<0. 05). Conclusions Xinfeng capsule can improve inflammation in patients with AS by the immune-mediated blood stasis, and the mechanisms could related to down-regulated expression of NF-κB and maintain body Th1/Th2 state of dynamic equilibrium.关键词
