氟对大鼠牙胚中TGF-β1和AMTN表达的影响

Abstract
Objective:To explore the effect of fluoride on the expression of TGF-β1 and AMTN in teeth germ.Meth-ods:SD rats teeth germs were cultured in vitro with Trowell's method.The tooth germ was exposed to serum medium contai-ning sodium fluoride at a concentration of 0,0.5,1.0,4.0 mmol/L,respectively.After exposure for 1,3,7 d,RT-qPCR tech-nique was used to observe mRNA relative expression level of TGF-β1 and AMTN mRNA.Results:Compared with the control group,the expression of TGF-β1 and AMTN in teeth germs of all exposure groups decreased significantly and showed an obvi -ous dosage and time dependent manner .The expression level of TGF-β1 in all exposure groups was significantly positively re-lated with it of AMTN,respectively.Conclusion:In the initial stage of enamel development ,the excessive fluoride inhibited the expression of TGF-β1 and AMTN in teeth germs ,which was an important pathological mechanism of mineralization of the enamel defects induced by fluoride .关键词
