PDGF-A基因修饰皮片对眼睑皮肤创伤修复的研究
PDGF-A gene modification in eyelid skin wound repair
摘要
Abstract
Objective To discuss PDGF-A gene modification in eyelid skin wound repair and the mechanism.Methods 6 minipigs were used in this study;3 round full-thickness wounds (φ3.67cm) were created on the dorsum of each side close to the vertebral column in each pig;in total,36 wounds were created;18 right side wounds were treated with skin sheet of PDGF-A gene modification as experimental group (group A),6 left side wounds were treated with human amniotic membrane (HAM) loaded withbonemarrow-derivedmesenchymalstemcells (bMSCs) and keratinocyte (group B) and 12 left side wounds were treated with HAM as control (group C);the granulation tissue,reepithelization and wound area were observed after 1,2,3 and 4 weeks;immunohistochemistry was performed using Laminin as a marker for blood vessels,as well as the detection of collagen content.Results The healing time of group A was 15 to 17 days,that of group B was 21 to 23 days and that of group C was 24 to 26 days;the healing time of group A was 5 to 6 days faster than that of group B and 8 to 9 days faster than that of group C;15 to 17 days after the graft,the granulation tissue,which contained a lot of Laminin,fibroblasts,capillaries and collagen,grew quite well in group A while many inflammatory cells were still seen in group B and group C,and the contents of Laminin,fibroblasts,capillaries and collagen in granulation tissue were smaller than those in group A.Conclusions The graft of skin sheet of PDGF-A gene modification is of good healing effect on eyelid skin wounds,the wounds heal with high quality.关键词
