摘要

Abstract

Objective To investigate the effects of stress and charpone (STCH) gene knock-out on neurological function in mice with MPTP-induced parkinson's disease (PD).Methods MPTP (30 mg/ml for 5 d) was injected intraperitoneally in STCH +/-mice to induce the experimental PD model.Rotarod test,forced swimming test,open field test,morries water maze behavioural test were conducted in experimental and control groups 1,2 and 3 week after MPTP injecteion.The animals were sacrificed and brain samples were collected 3 week after MPTP injection.The mRNA experssion levels of TNF-α,IL-1β,IL-10 and NGF were tested by RT-PCR;the neurotransmitters dopamine and 5-serotonin levels were detected by HPLC-MS.Results Motor and coordination abilites results did not display significant differences between the MPTP + STCH (+ /-) and STCH (+ /-) mice at 1 or 2 week after modeling.But depression appeared in MPTP + STCH (+/-) mice during these time points.MPTP + STCH (+/-) mice appeared obvious motor disablities 3 week after modeling,while there were no significant differences in learing and coordination ablities between two groups.There were no significant differences in the expression of inflammatory factors and the contents of dopamine and 5-HT in brain tissue between the MPTP + STCH (+/-) and STCH (+/-) mice.Conclusion STCH gene knock-out may cause depression at early stage of MPTP-induced PD model,but may not affact motor and learning abilities.
