太白楤木对肝星状细胞凋亡及相关基因表达的影响
Effects ofAralia on apoptosis and expression of apoptosis-related genes in hepatic stellate cells

Abstract
Objective To investigate the effects of Aralia on apoptosis and expression of apoptosis-related genes in hepatic stellate cells (HSC-T6 )and to explore the anti-fibrotic mechanism of Aralia.Methods We divided 30 male SD rats at random into negative control group,colchicine-treated and aralia-treated groups,with 10 in each.After administered with normal saline,colchicine and aralia by gavage separately for 3 consecutive days, serum containing the drug was prepared.Then HSC-T6 cells were treated with serum containing the drug mentioned above for 48 h.Flow cytometry was used to assess apoptosis.Reverse transcription PCR (RT-PCR)was used to detect mRNA expressions of Bcl-2 ,Bcl-xs,Bax and Bcl-xl.And the protein expressions of Bcl-2 ,Bax andα-SMA were determined by Western blot.Results ① The apoptosis rates of HSCs in colchicine-treated and Aralia-treated groups were significantly higher than that in negative control group (P<0 .05 );Aralia-treated group had a higher apoptosis rate than colchicine-treated group (P<0 .05 ).② Colchicine and Aralia could decrease Bcl-2 expression, but increase Bax expression markedly (P<0 .05 );and Aralia exerted a more obvious effect.③ There was no significant difference in the mRNA expressions of Bcl-xs and Bcl-xl among the three groups.④ Colchicine and Aralia could inhibitα-SMA expression evidently (P<0.05),and the effect was more obvious in Aralia-treated group.Conclusion Aralia can significantly accelerate the apoptosis of HSC-T6 cells possibly by up-regulating Bax expression and down-regulating Bcl-2 expression.关键词
