Abstract

Objective:To detect the expression of histone demethylases KDM5 B ( JAR-ID1B/PLU-1) in ovarian cancer tissue and cell line,to analysis its effect on ovarian cancer chemoresistance. Methods:Detect the expression of KDM5B in 36 cases of epithelial ovarian cancer,15 cases of normal epithelial ovarian tissue,6 pairs of matched primary surgical resec-tion specimens and recurrent specimens,ovary normal epithelial cell ( IOSE) and five ovarian cancer cells by quantitative reverse transcriptase PCR ( RT-qPCR ) . Lentiviral expression and RNAi vectors were constructed and obtained stably overexpressing SKOV3 and interference Ca-ov3 cell lines. RT-qPCR and Western blot was used to identify the effect of regulation. The sta-ble overexpression and knockdown of KDM5 B cell lines were treated with different concentrate of cisplatin,then CCK-8 were used to assay cell viability and calculate the cisplatin half inhibi-tory concentration (IC50). Results:The average value of KDM5B mRNA transcript in normal o-vary tissue was lower than that in ovarian cancer(1. 950±0. 3003 vs 4. 924±0. 2989;t=5. 909, P<0. 0001) and up-regulated in five of six recurrent patients. KDM5B expression was signifi-cantly correlated with FIGO stage,pathological grade,metastasis and drug resistance of ovarian cancer patients,which was not related to the age of the patients and the level of serum CA125. Compared with IOSE cells,the KDM5B expression were increased gradually in SKOV-3、HO-8910、ES-2、 A2780、Caov3 cell lines. RT-qPCR results showed that KDM5B overexpression power was 27 . 4 times in SKOV3 cell lines and knockdown effect were 0 . 38 and 0 . 41 in the sh1 and sh2 groups in Caov3 cell lines. Western blot also showed that the expression and inter-ference were effective. The KDM5B mRNA and protein expression were gradually increased in Caov3 cell line when treated with increasing concentration gradients of cisplatin,IC50 of KDM5B over expression SKOV3 cell line[3. 666(95%CI:3. 067~4. 382) μg/ml]was higher than that of the control group[1. 676(95%CI:1. 553~1. 810) μg/ml ]and Caov3 cell line interference sh1 group[3. 359(95%CI:2. 875~3. 925) μg/ ml],sh2 group was 2. 881(95%CI:2. 49~3 . 324 ) μg/ml lower than that in the control group [ 6 . 972 ( 95%CI:6 . 182~7 . 864 ) ] . Con-clusion:Histone demethylases KDM5B is overexpressed in ovarian cancer, and promotes the chemoresistance of ovarian cancer cell line.
