鸡血藤总黄酮对脂多糖刺激RAW264.7细胞氧化应激的调节作用
Effect of total flavonoids of Spatholobus suberectus Dunn on LPS-induced oxidative stress in RAW264.7 cells

Abstract
Total flavonoids of Spatholobus suberectus Dunn (FSSD) was obtained by ethanol extraction.The cytotoxicity of FSSD was analyzed using MTT assay.Its anti-oxidative stress activity was evaluated by both pre-treatment and post-treatment of RAW264.7 cells under oxidative stress induced by lipopolysacchride (LPS) with FSSD.For pre-treatment,RAW264.7 cells were treated with series concentration (25,50 and 100 μg/mL) of FSSD for 12 h before LPS stimulation.For post-treatment,RAW264.7 cells were firstly stimulated by LPS for 2 h and then treated with series concentration of FSSD for 8 h.NO and reactive oxygen species (ROS) levels were analyzed using Griess assay and fluorescent probe label method.It was found that both FSSD pre-treatment and post-treatment can inhibit the production of NO and ROS induced by LPS,and post-treatment showed better effect compared to pre-treatment method.Thus,FSSD has high anti-oxidative stress activity and it is more suitable to be used for treatment of oxidative stress rather than prevention.关键词
