Abstract

The protective effects of Kisspeptin in rats after heat stress were investigated using metabonomic method.Gas chromatography coupled to time-of-flight mass spectrometry (GC/TOFMS) was used to detect the serum endogenous metabolites in rats.Serum metabolite profiles were analyzed with multivariate statistical analysis,such as principal component analysis (PCA) and partial least squares-discriminate analysis (PLS-DA).The result showed that a clear difference was observed among control,heat stress and heat stress treated with Kisspeptin groups.The differentially expressed metabolites and potential biomarkers were selected and identified according to parameters of variable importance in the projection (VIP) and one-way ANOVA.Results showed that the serum metabolic profiling of heat stress-induced rats was significantly different from that of control group,and it had the tendency to the normal state after Kisspeptin treatment.The metabolic disorders of TCA cycle,saccharometabolism,amino acid,and nucleoside metabolism were induced by heat stress based on biochemical metabolic pathway analysis.However,the metabolic disorders were rescued after Kisspeptin treatment.The oxidative stress parameters analysis showed that Kisspeptin treatment significantly inhibited malondialdehyde (MDA) level,and restored superoxide dismutase (SOD),total antioxidant capacity (T-AOC) and glutathione peroxidase (GSH-Px) activities.This study provides the experimental basis for healthy breeding and the development of safe biological products for anti-heat stress.
