快速检测HPV16的环介导等温扩增检测法的建立
Establishment of loop-mediated isothermal amplification assay for rapid detection of HPV16
摘要
Abstract
Objective To establish a method of loop-mediated isothermal amplification assay (LAMP) for rapid detection of human papillomavirus 16 (HPV16).Methods Primers were designed for HPV16 E7 to establish the LAMP reaction system,of which the detection sensitivity was verified,and comparisons with PCR method,LAMP method using loop primer,and LAMP kit method were made.Results The genes in E7 of HPV16 virus can be detected successfully by use of LAMP method.The detection can be completed after the reaction under an isothermal condition for 60 min.After loop primer was added,the sensitivity of LAMP method was higher than that of PCR method and equivalent to that of LAMP kit method,i.e.,a dilution multiple of 103.Conclusion LAMP method has the advantages of high sensitivity,simple and convenient operation,no need of complex special instruments,and it is applicalbe for the rapid detection of HPV16 virus,so it is worthy of development and application.关键词
