肥厚型心肌病诱导多能干细胞模型的建立
Establishment of induced pluripotent stem cell model of hypertrophic cardiomyopathy

Abstract
Objective To establish patient-specific induced pluripotent stem cell model of hypertrophic cardiomyopathy (HCM). Methods Peripheral blood mononuclear cells (PBMCs) were isolated and cultured from whole blood of patients with HCM. And then, episomal combinations coding Sox2, Klf4, Oct4, L-Myc, Lin28, mp53DD, EBNA1 were electroporated into PBMCs. Pluripotency and stability of the induced pluripotent stem cells (iPSCs) were tested by standard procedures including cell morphology, alkaline phosphatase staining, pluripotency markers staining, and potential differentiation of all three germ layers was detected through em-bryonic body (EB) formation test. Results The patient-specific iPSCs possessed the characteristic of human embryonic stem cells (ESC), confirmed by methods including clearly ESC-like colonies, alkaline phosphatase staining, immunofluorescence and RT-PCR for expression of pluripotency markers, which were similar to hu-man ESC H9. Additionally, EB formation test demonstrated that iPSCs hold potential to differentiate into all three germ layers. Conclusions Hypertrophic cardiomyopathy patient-specific iPSCs are successfully obtained, which lays the foundation of further construction of induced cardiomyocytes HCM disease model, mechanism study and drug discovery.关键词
