Abstract
Objective To establish an HPLC limit detection method for douber-ester alkaloids in Yanghe plaster. Methods The sep-aration was performed on Inertsil ODS-3 column ( 250 mm × 4. 6 mm, 5 μm ) . The mobile phase were acetonitrile-tetrahydrofuran ( 25:15 ) and 0. 1 mol/L ammonium acetate-water with gradient elution. The flow rate was 1 mL/min with column temperature 20 ℃ and detec-tion wavelength at 235 nm. Results The calibration curves were within the ranges of 0. 2148-2. 1480 μg for hypaconitine, 0. 1968~1. 9680 μg for mesacontitine and 0. 2040-2. 0400 μg for aconitine. The average recoveries were 93. 24%, 98. 49% and 94. 93%, RSD were 2. 12%, 2. 51% and 2. 65%( n=6 ) , respectively. Conclusion This method is reliable, reproducible, which can be applied to quantitative analysis of this preparation.关键词
