Abstract
Objective To investigate the expression of connective tissue growth factor (CTGF) in kidney tissue of rat hydronephrosis model.Methods Forty eight healthy male Wistar rats were equally divided into two groups:in model group the left pelviureteric junction was dissociated and the ureter was ligated,and in control group the left pelviureteric junction was dissociated only.Six rats in each group were sacrificed 1,2,4 and 8 weeks after the operation and the left kidney was taken for histopathological examination.The expression of CTGF protein and mRNA in kidney tissue was detected with immunohistochemistry and Western blot,respectively.Results The expression of CTGF mRNA and protein in model group was significantly higher than that in the control group (P<0.01) and increased with the time length of pelviureteric junction obstruction.Conclusion The expression of CTGF is upregulated in the rat hydronephrosis model and positively correlated with the obstruction duration,indicating that CTGF might be used as a target of preventing congenital hydronephrosis.关键词
