Effect of solid-state cultural conditions on biosynthesis of secondary metabolites in taxol-producing fungus P.microspora

摘要

Abstract

Paclitaxel is widely used in the treatment of solid tumors,but its production remains short of demand in the global market.Fermentation of entophytic taxol-producing fungi may solve the shortage issue.However,the taxol yield of production in the laboratory remains low.Solid-state fermentation (SSF) for the production of fungal secondary metabolites has drawn much attention in recent years due to a number of advantages over liquid culture techniques.We in this study used SSF to search for taxol-like molecules in taxolproducing fungus Pestalotiopsis microspora.It was found that in SSF the fungus produced two additional compounds that were absent in liquid-state fermentation.One compound was purified by HPLC with a retention time similar to taxol standard.A maximum yield of 0.354 mg per gram of cultivating matrix was achieved.Therefore change of cultivation method can induce further secondary metabolites in fungus.Culture media and time in solid-state fermentation for P.microspora were also tested.
