Abstract

The objective of this study was to estimate the influence of different inorganic phosphorus level diets supplemented with phytase on bone calcium and phosphorus metabolism of weaned piglets.A total of 192 healthy post-weaned piglets (Duroc×Landrace×Largewhite) with an average body weight of (9.12±0.29) kg were randomly divided into 6 groups with 4 replicates in each group,and 8 piglets per replicate.Control group was fed a basal diet (phosphorus content was 6.00 g/kg),experimental groups were fed diets supplemented with 750 U/kg phytase,and inorganic phosphorus addition level was 100%,75%,50%,25% and 0 (calcium-phosphorus ratio was 1.17 to 1.79) of the control group,respectively.The experiment lasted for 35 d.The results showed that with phytase supplementation,the dietary inorganic phosphorus level reduction resulted in a linear and quadratic decrease in contents of tibia and metatarsal bone calcium,phosphorus and ash and the rib bone phosphorus content (P<0.05),and got the same effects on contents of serum fibroblast growth factors (FGF23),biochemical indices of bone synthesis such as bone gla protein (BGP) and osteoprotegerin (OPG) contents,and bone-specific alkaline phosphatase (BAP) activity (P<0.05);it also resulted in a linear and quadratic increase in serum calcitonin (CT) content,biochemical indices of bone resorption such as contents of C-terminal telopeptide of type Ⅰ collagen (CTX-Ⅰ) and osteopontin (OPN),and tartrate-resistant acid phosphatase (TRAP5b) activity,as well as 1,25-dihydroxyvitamin D [1,25-(OH) 2D3] activity (P<0.05).Compared with control group,75% inorganic phosphorus level + phytase group had no significant difference on contents of bone calcium,phosphorus and ash,and biochemical indices of bone synthesis and resorption (P> 0.05);50% inorganic phosphorus level+phytase group had no significant difference on bone ash content and biochemical indices of bone synthesis (P>0.05),but got significantly higher biochemical indices of bone resorption (P<0.05).Meanwhile,these two groups above both had significantly higher serum 1,25-(OH) 2D3 activity and CT content (P<0.05),but a significantly lower FGF23 content compared with control group (P< 0.05).In summary,with phytase supplementation in diets,contents of calcium,phosphorus and ash in tibias and metatarsus and phosphorus in ribs are linear and quadratic correlated with inorganic phosphorus addition level.Hormones and biochemical indices of bone metabolism can accurately reflect the situation of bone metabolism,and are more sensitive than conventional indicators.Dietary phytase supplementation for weaned piglets can replace 25% (0.52 g/kg) inorganic phosphorus.
