摘要

Abstract

Objective To investigate injury effect of high glucose concentration on original generation of human umbilical vein endothelial cells (HUVECs) and its injury mechanism.Methods HUVECs were cultured by medium of low concentration of glucose (LG,5.5 mmol/L) and high concentration of glucose (HG,35 mmoL/L) respectively for 24 h,72 h and 120 h.Effect of HG on survival rate of HUVECs was detected by using cholecystokinin-8 (CCK-8) assay,and reactive oxygen species (ROS) and nitric oxide (NO) levels were detected using fluorescent probe,and then expressions of HG on Bax,Bcl-2,silent mating type information regulation 2 homolog 1 (SIRT1) and endothelial nitric oxide synthase (eNOS) expressions were detected by western bolt method.Results In HG group,after culturing for 72 h and 120 h,values of survival rate and Bcl-2 were lower,while values of ROS and Bax/Bcl-2 ratio were higher than those in LG group (P < 0.05);after culturing for 24 h,72 h and 120 h,relative amounts of SIRT1,and eNOS were lower than those in LG group;after culturing for 72 h and 120 h,eNOS expressions were lower than those in LG group (P < 0.05).Conclusion HG may decrease survival rate,increase of intracellular oxidative stress reaction,improve Bax/Bcl-2 ratio of HUVECs,which may be associated with inhibition of SIRT1-eNOS pathway to reduce NO production.
