组蛋白甲基转移酶对人脑胶质瘤细胞迁移和侵袭的影响及其机制的研究
Effect and mechanism of the SET and MYND domain-containing protein 2 (SMYD2) on cell migration and invasion of human brain glioma cells

Abstract
Objective To investigate the expression of the SET and MYND domain-containing protein 2 (SMYD2) in glioma and the mechanism on cell migration and invasion of human brain glioma U87,U251 cells.Methods Expression of SMYD2 in 50 cases of glioma tissues and 10 cases normal brain(NB) tissues were analyzed by immunohistochemistry.Small interfering RNA(siRNA) was used to transfect U87,U251 human glioma cells respectively,the expression levels of SMYD2 and matrix metallopeptidase-9 (MMP-9), p-p65,human tissue inhibitor of metalloproteinases-1 (TIMP-1) proteins in human brain glioma U87,U251 cells were detected after SMYD2 knockdown by using Western blotting.The migration and invasion ability of U87,U251 cells were respectively measured by wound-healing assay and transwell migration and invasion assays.Results Our results showed that SMYD2 expression gradually increased in normal brain,gradesⅠ-Ⅱ and grades Ⅲ-Ⅳtumor(P<0.05).Meanwhile, a positive relationship between the level of SMYD2 and the malignancy of glioma was determined(r=0.635,P<0.05).The expression of SMYD2 was decreased obviously in SMYD2-siRNA group and the expression of MMP9, p-p65 were down-regulated and TIMP1 were up-regulated respectively compared with the Negative Control-siRNA group(P<0.05-0.01).The migration ability of glioma cells was decreased obviously in wound-healing assay after SMYD2 knockdown (all P<0.01).The result of transwell assay indicated that cell migration and invasion ability of SMYD2-siRNA group were decreased obviously(all P<0.05).Conclusion The expression level of SMYD2 was positively associated with the malignancy of glioma.SMYD2-targeted siRNA interferes with the expression levels of SMYD2,and down-regulated the expression of smyd2 can suppress the migration and invasion of glioma U87,U251 cells obviously.关键词
