Abstract

Objective To optimize the extraction technology for the self-made of Chinese medicine and investigate the effect of the extraction on learning-memory ability in model mice induced by Scopolamine.Methods The dry extract yield was chosen as index.The four factors of the extraction solvent and time,the solvent ratio,and soaking time were investigated by L9(34) orthogonal test.Kunming mice were randomly divided into normal control group,scopolamine model group,positive control drug,low,medium and high doses of the self-made of Chinese medicine(0.5,1.0 and 2.0 g·kg-1).The drugs were intragastrically administered to mice for four weeks.On day 28,after intraperitoneal injection of scopolamine,mice were subjected to Morris water maze tests.Results The optimal technology was added to 12 times the amount of 50% ethanol,soaking 90 min,refluxing and extracted three times,each extraction for 2 hours.In animal experiments a significant decrease of swimming time in Morris water maze test was observed in the self-made of Chinese medicine groups(P<0.05).Conclusion The optimal technology for the self-made compound extract of Chinese medicine was reasonable,the extraction from which could effectively improve learning-memory ability in model mice induced by scopolamine.
