Abstract
Objective To investigate the expression of monocyte chemoattractant protein-1 (MCP-1) in human adipose tissues and its significance.Methods Epiploic fatty tissues were collected from patients with cholecystolithiasis for selective surgery.The mRNA expression of tumor necrosis factor-α (TNF-α), MCP-1, and toll-like receptor 4 (TLR4) was measured by real-time fluorescence quantitative RT-PCR.The regression analysis was used to examine the relationship between the expression levels of these factors and body mass index (BMI).Preadipocytes were isolated from epiploic fatty tissues in patients with cholecystolithiasis for selective surgery and were cultured in medium.Cell morphological changes were observed under the microscope.Cells were randomly divided into two groups: the observation group was given a final concentration of 500 g/ml free fatty acid (FFA) for the treatment, and the control group was cultured with primary medium.Twenty-four hours after treatment, the relative mRNA expression of TNF-α, MCP-1, and TLR4 was measured by using real-time fluorescence quantitative RT-PCR.Results The mRNA expression levels of MCP-1, TNF-a, and TLR4 were 1.20±0.07, 0.57±0.07, and 1.22±0.08, respectively.The expression of MCP-1 in adipose tissues was positively correlated with BMI (r2=0.2343, P<0.05);the expression of TNF-α was also positively correlated with BMI (r2=0.428 6, P<0.01).The expression of MCP-1 (P<0.05) and TNF-α (P<0.05) increased after FFA treatment.No significant difference was found in the expression of TLR4 between these two groups (P>0.05).Conclusion The expression of MCP-1 in adipose tissues increases with the increase of BMI, and the MCP-1 expression is increased and inducing obesity may be caused by regulating TLR4 signaling pathway and promoting the release of TNF-α.关键词
