EGCG对尿酸诱导人脐静脉内皮细胞炎症反应及氧化应激的影响
Effect of EGCG on inflammation and oxidative stress in uric acid-induced HUEVCs

Abstract
Objective To investigate the effect of epigallocatechin gallate (EGCG) on the inflammation and oxidative stress (OS) in uric acid (UA)-induced human umbilical vein endothelial cells (HUVECs).Methods HUVECs were cultured in vitro and cells of the 4th-5th passage were divided into four groups.The control group was cultured with EGM-2 complete medium;UA group was added with EGM-2 complete culture medium with the final concentration of 8 mg/dL UA;UA+EGCG group was added with EGM-2 complete culture medium with final concentration of 50 μmol/L EGCG for 12 h, and then the final concentration of 8 mg/dL U was added into the EGM-2 complete medium;and EGCG group was added with EGM-2 complete medium with the final concentration of 50 μmol/L EGCG.The cells were collected at 24 hours.RT-PCR was used to detect the mRNA expression of NF-κB, monocyte chemoattractant protein-1 (MCP-1), intercellular adhesion molecule-1 (ICAM-1), cyclooxygenase 2 (COX-2), TNF-α and inducible nitric oxide synthasel (iNOS).The level of reactive oxygen species (ROS) was determined by DCFH-DA method.Results The relative expression of NF-κB, MCP-1, ICAM-1, TNF-α and iNOS mRNA in the UA group was significantly higher than that in the control group (all P<0.01).The mRNA expression of NF-κB P65, MCP-1, ICAM-1, COX-2, TNF-α and iNOS in the UA+EGCG group and EGCG group was significantly reduced as compared with that of the UA group (all P<0.01).The mean fluorescence intensity of ROS in the UA group was significantly higher than that in the control group (345.98±27.01 vs 101.26±12.34), The mean fluorescence intensity of UA+EGCG group was 112.65±9.62, which was significantly lower than that of UA group (all P<0.01).Conclusion EGCG can attenuate the inflammation and OS of UA-induced HUVECs by inhibiting NF-кB signal pathway and reducing the release of inflammatory factors.关键词
