PD1/PDL1信号途径在小鼠变应性鼻炎发病中的作用
Effects of PD1/PDL1 signal pathway in allergic rhinitis of mice

Abstract
Objective To investigate the effects of PD1/PDL1 signal pathway in allergic rhinitis (AR) of a murine model.Methods Twenty-four BALB/c mice were randomly divided into four groups, six animals in each.The mice in the model group, PDL1 monoclonal antibody (anti-PDL1 mAbs) group and PDL1 recombinant protein group (PDL1 group) received ovalbumin (OVA) to reduce allergic rhinitis.PDL1 monoclonal antibody or recombinant PDL1 protein was administered intranasally to mice in the latter two groups.Mice in the control group were administrated normal saline.Nasal rubbing motion, sneezing and rhinorrhea were evaluated by superposition quantization scoring.The positive cells and integrated optical density of PD1 and PDL1 in nasal mucosa was analyzed by immunohistochemistry.The concentration of TGF-β, IL-10 and IL-17 in serum was detected by ELISA.The expression of Foxp3 and RORγt in nasal mucosa was measured by Western blotting.Results The behavioral scores for the control, model, anti-PDL1 mAbs and PDL1 groups were 0, (6.17±0.98), (7.33±0.82) and (3.17±1.17) points, respectively (all P<0.01).Significantly decreased concentration of TGF-β, IL-10 in serum, and Foxp3 in nasal mucosa was observed in model group and anti-PDL1 mAbs group as compared with that of the control group (P<0.01), while significantly increased expression of IL-17 in serum and RORγt in nasal mucosa was found (P<0.01).The same trend was observed for the anti-PDL1 mAbs group as compared with the model group (P<0.01), and the opposite trends for PDL1 group as compared with the model group (P<0.05 or P<0.01).Conclusion PD1/PDL1 signal pathway is involved in pathogenesis of AR.PDL1 may inhibited AR by regulating Treg/Th17 balance and promoting the differentiation of Treg cells.关键词
