急性坏死性胰腺炎大鼠水通道蛋白4与脑损伤关系探讨
Study on relationship between aquaporin-4 and cerebral injury in rats with acute necrotizing pancreatitis

Abstract
Objective To explore the change in aquaporin-4 (AQP4) in the cerebral tissue of the rats with acute necrotizing pancreatitis (ANP) and analyze the association of AQP4 with blood-brain barrier permeability and brain edema. Methods Eighty-four adult male SD rats were divided into the ANP groups (n=72) and sham operation group (n=12). The rats with ANP were designated 3, 6, 9, 12, 18 and 24 h after pancreatitis model induced by injecting sodium taurocholate. The brain water content in rats and the blood-brain barrier permeability at each time point were measured respectively. At the same time, the expression of AQP4 both protein and mRNA in brain tissue was detected with Western blot and semi-quantitative reverse transcription polymerase chain reaction. Results The expression of AQP4 protein and mRNA in the brain tissue was significantly up-regulated at 6 h and increased at 9, 12, 18 and 24 h successively (P<0.05). It was shown that the expression of AQP4 protein and mRNA was positively correlated with the brain water content (P<0.05). Conclusions There would be decrease in blood-brain barrier function and increase in the expression of protein and mRNA of AQP4 in the brain tissue of rats with ANP, which may be closely related to the formation and development of cerebral edema.关键词
