摘要

Abstract

Objective To investigate the effect of hemodilution with succinylated gelatin (GEL), hydroxyethyl starch (HES) 130/0.4 and lactate Ringer solution (RL) on thrombin generation. Methods Blood samples were obtained from 8 healthy adult volunteers. Each blood sample was diluted with GEL (group GEL), HES130/0.4 (group HES) and RL (group RL). The dilution ratios of citrated blood volume to plasma substitute volume were 10:0 (undiluted), 10:2, 10:4 and 10:6. Coagulation factorⅡactivity (FⅡ:C) and antithrombin activity (AT:A) were detected by individual assays and thrombin generation was detcted by calibrated automated thrombography (CAT). Thrombin generation curve using CAT showed the five main parameters of lag time, time to peak (ttpeak), peak thrombin (peak), start tail and endogenous thrombin potential (ETP). Results FⅡ:C decreased obviously when dilution ratios increased in each group compared to the baseline value, but showing no significant differences between groups. Ttpeak increased and peak decreased in group GEL and group HES, and lag time was prolonged at extensive dilution of HES, showing significant differences from group RL. Neverthe-less, ETP was not significantly changed in all groups. AT:A decreased after dilution with GEL and HES when compared with RL. Conclusions HES130/0.4 and GEL decrease velocity and peak of thrombin, additionally, HES130/0.4 delays lag time of thrombin. Both reduce antithrombin activity. RL contributes to maintain normal blood coagulation function.
