摘要

Abstract

Objective To investigate the effects of paclitaxel on levels of cluster of differentiation 28 (CD28),cytotoxic T lymphocyte-antigen-4 (CTLA-4),transforming growth factor beta (TGF-β) in experimental autoimmune encephalomyelitis (EAE) of rats.Methods The 50 rats were divided into 5 groups by the random number table:the doses of small,middle,large of experimental groups were intraperitoneal injection of 1,2,4 mg · kg-1 paclitaxel for 10 consecutive days;the normal group and model group were injected 0.9% NaC1 2 mL.Each group had ten rats.The levels of CD28 and CTLA-4 were detected by flow cytometry.The levels of TGF-β in blood were measured by radiaoimmunoassay.Results The nerve dysfunction score in small,middle,large doses experimental groups were (2.30 ± 1.16),(1.10 ±0.57),(0.90 ±0.57) point.That was lower than model group (3.90 ± 0.99) point.The comparative differences between groups were statistically significant (P < 0.01).The brain tissue score in small,middle,large doses experimental groups were (1.61 ± 0.24),(91.44 ± 0.22),(1.23 ± 0.31) point.That was lower than model group (2.67 ± 0.52) point.The comparative differences between groups were statistically significant (P < 0.01).The expression of CD28 in small,middle,large doses experimental groups were 33.9 ± 4.49,26.4 ± 4.14,20.7 ± 4.21.That was lower than model group 41.2 ± 4.09.The comparative differences between groups were statistically significant(P < 0.01).The expression of CTLA-4 in small,middle,large doses experimental groups were 4.3 ± 1.42,5.8 ± 1.81,6.4 ± 1.68.That was higher than model group 3.6 ± 1.32.The comparative differences between groups were statistically significant (P < 0.01).The levels of TGF-13 in small,middle,large doses experimental groups were (11.20 ± 1.80),(11.30 ± 1.83),(14.10 ± 1.87)ng· mL-1.That was higher than model group(11.00 ± 1.73) ng · mL-1.The comparative differences between groups were statistically significant (P < 0.01).Conclusion PTX could decrease the nerve dysfunction score and brain tissue score,the mechanism may adjust the expression of CD28,CTLA-4 in brain and the expression of TGF-β3 in serum.PTX may have preventive and therapeutic effects on EAE rats.
