注射用血栓通及其有效组分对缺氧/复氧损伤神经元保护作用
Protective effects of Xueshuantong injection and its effective constituents on neurons with hypoxia/reoxygenation injury

Abstract
Objective To compare the protective effects of Xueshuantong injection and its effective constituents on neurons cells with hypoxia/ reoxygenation(HL/R) injury.Methods Neuro-2a cells were cultured and divided into control group,model group,administration group (Panaxadiolsaponin group,Panaxatriolsaponin group and Xueshuantong group) and compatibility group.The model of H/R was established.The H/R cells were treated with different concentrations of Panaxadiolsaponins,Panaxtrolsaponins and Xueshuantong (50,100,200,400 and 800 mg · L-1) for 24 h.The cells in the compatibility group of Panaxadiolsaponins,and Panaxtrolsaponins(100 mg · L-1) were treated for 24 h.The cell viability and the leakage of lactate dehydrogenase (LDH) in each group were detected by CCK-8 and the leakage of lactate dehydrogenase (LDH) Kit method.Results Compared to model group on the survival rate of cells with (51.13 ± 1.74) %,the survival rates of cells with (24.27 ±4.88)%,(1.13 ± 0.3)%,(0.93 ±0.30)% in Panaxadiolsaponin group at the concentrations of 200,400,800 mg · L-1 were decreased(P <0.01).Compared to model group on the leakage rate of LDH with (260.97 ± 10.86)%,the leakage rates of LDH with (319.33 ± 31.37)%,(599.32 ± 34.11)%,(879.58 ±47.20)% in Panaxadiolsaponin group at the three concentrations were increased(P <0.01).Compared to model group on the survival rate of cells with (51.16 ± 6.32)% and the leakage rate of LDH with (361.20 ± 26.43) %,the survival rates of cells with (66.26 ± 3.15) %,(60.23 ± 1.93) %,(62.22 ± 4.79) % were increased and the leakage rates of LDH with (295.97 ± 23.62) %,(235.97 ± 43.80) %,(173.33 ± 26.53) % were decreased in Panaxtrolsaponin group at the three concentrations (P < 0.01).Compared to model group on the survival rate of cells with (50.01 ±2.83)% and the leakage rate of LDH with(270 ±74.24)%,the survival rates of cells with (58.63 ± 6.07) %,(65.25 ± 8.80) %,(66.38 ± 8.62) % were increased and the leakage rates of LDH with (171.45 ± 13.69) %,(190.55 ± 17.90) %,(187.07 ± 30.09) % were decreased in Xueshuantong group at the concentrations of 100,200,400 mg · L-1 (P < 0.01).Compared to model group on the survival rate of cells with (37.29 ± 3.73) %,the survival rates of cells with (46.57 ± 1.27) %,(43.29 ± 2.29) % in the second and fifth compatibility group were increased and the cell survival rate with (27.92 ± 2.89)% was decreased in the fourth compatibility group significantly (P < 0.05,P < 0.01).Compared to model group on the leakage rate of LDH with (252.67 ± 9.35) %,the rates of LDH leakage with (211.17 ± 7.73) %,(227.17 ± 13.91) % in the second and fifth compatibility group were decreased and the rate of LDH leakage with (295.83 ± 12.77) % was significantly increased in the fourth compatibility group (P < 0.05,P < 0.01).Conclusion Xueshuantong injection and its effective components had a protective effect on neurons cells with H/R injury.关键词
