秋水仙素对大蒜愈伤组织四倍体的诱导效应及试管鳞茎快繁体系构建
Tetraploid Inductive Effect of Colchicine in Garlic Callus and Construction of Rapid Propagation System of Bulblets in vitro

Abstract
Taking garlic cultivar'Liubanhong'bulb pies as explants to induce callus,and colchicine as mutagenic agent,this paper conducted polyploid induction on the callus and compared the effect of different colchicine concentrations and treating time on garlic callus. The results showed that the tetraploid inducing rate was increased along with the increasing of colchicine concentration,while showing a tendency of first increasing then decreasing along with the increase of colchicine treatment time. The chromosomal aberration rate was increased along with the increasing of colchicine concentration and treatment time. When the concentration of colchicine was 0.200%,the treating time was 18 h,the highest tetraploid inductivity was 36.36%,which could be used as the optimum induction system. The callus treated by colchicine could successfully regenerate test-tube plantlet and then bulblet in vitro was gained.关键词
