抗菌肽CGA-N12对腹腔注射热带念珠菌小鼠的治疗作用
Curative Effect of Antifungal Peptide CGA-N12 on Mice Systemically Infected with Candida tropicalis

Abstract
CGA-N12,a derived peptide of chromogranin A which consists of the 65th to 76th amino acids,has specific antifungal activity,especially to clinical pathogenic fungus Candida tropicalis.The study was designed to research the antifungal activity of CGA-N12 in vivo.The mouse model systemically infected with Candida tropicalis was established by injecting clinical isolated strain of Candida tropicalis via intraperitoneal.15 mg/kg and 30 mg/kg CGA-N12 were applied for treatment,respectively.We set 15 mg/kg itraconazole-treated mice as therapeutic control group and PBS-treated mice as infected control group.The survival rate,the changes of body weights and the Candidas-loading amount of kidney were calculated;pathological lesions of tissues and organ with HE staining were observed.The results showed that,compared with control group,after treatment with 15 mg/kg and 30 mg/kg CGA-N12 for 14 d,the Candidas-loading amount of kidney reduced 97.6%and 99.1%,the survival rate increased 40%and 45%,the average body weight increased 1.3 g and 5.9 g,and the immune organ indexes increased significantly,respectively.The difference of the results between 30 mg/kg CGA-N12 and 15mg/kg/d itraconazole was not significant.The results of pathological lesions showed that 15 mg/kg and 30 mg/kg CGA-N12 both had therapeutic activity on the organ lesions of the infected mice and had no proliferation of hepatic vessel endothelial cell hyperplasia and renal hemorrhage which were found in itraconazole treated group.In conclusion,CGA-N12 has therapeutic activity on the mice systemically infected with Candida tropicalis.关键词
