NOX4介导缺氧诱导A549细胞侵袭的机制研究
The mechanism of NOX4 on hypoxia-induced A549 cells invasion

Abstract
Objective:To investigate the mechanism of NADPH oxidase 4 (NOX4) on hypoxia-induced A549 cells invasion.Methods:A549 cells were divided into an ormoxia group and a hypoxia group.The expression of HIF-1α mRNA in A549 cells was determined by RT-PCR.The invasion of A549 cells were detected by trans-well invasion assay.The protein expressions of NOX4,Akt and phospho-Akt (p-Akt) were detected by western blotting.Results:Compared with the normoxia group,the invasion of A549 cells,the ROS level,the HIF-1α mRNA expression and the HIF-1α,NOX4 and p-Akt protein expression in hypoxia group were markedly increased (P <0.01).However,hypoxia-induced invasion was blocked by treated with the NOX4 inhibiter GKT137831 (25 μmol/L) and DPI (10μmol/L) or PI3K/Akt inhibitor LY294002 (30 μmol/L).Conclusion:NOX4 could contribute to the hypoxia-induced A549 cells invasion,possibly via the activation of PI3K/Akt signaling pathway.关键词
