人脐带间充质干细胞培养体系的优化
Optimization of human umbilical cord mesenchymal stem cells culture system
摘要
Abstract
Objective:To optimize human umbilical cord mesenchymal stem cells (hUCMSCs) by comparing the influence of LG-DMEM,DMEM/F12 and α-MEM culture mediumn.Methods:The hUCMSCs were derived from ambilical cord by tissue explant culture method,and maintained in different cell culture media (LG-DMEM,α-MEM and DMEM/F12).The expression of hUCMSCs surface markers was detected by flow cytometry.The adipogenic and osteogenic differentiation abilities were analyzed by oil redO staining and alizarin red staining,respectively.The migration and proliferation abilities of cells were compared among the three groups.Results:The surface markers and differentiation ability of cultured cells were in agreement with hUCMSCs characteristics.The spread of cells grown in α-MEM medium was superior to those in LG-DMEM and DMEM/F12 media.However,the cells grown in DMEM/F12 medium had best proliferation ability (P ＜0.05).Conclusion:hUCMSCs grown in α-MEM culture media lead to better spread out,whereas the proliferation of the cells was accelerated by using DMEM/F12 media.关键词
