金针菇中总多酚含量测定方法的优选
Optimization on determination of total polyphenol in Flammulina velutipes

Abstract
To optimize the conditions to determinate the total polyphenol in Flammulina velutipeswith validation results and other evaluating indicators,the applicability of Folin-Ciocalteu method,UV spectrophotometry and Prussian blue method was investigated,and the method for determination of total polyphenol in Flammulina velutipes was selected.With the absorbance as indicator,single-factor experiments of chromogenic conditions were studied,and the chromogenic conditions were optimized by quadratic orthogonal rotation combination design.Results showed that comparing the interference,sensitivity and stability of the three methods,Folin-Ciocalteu method was the optimal one for the determination of total polyphenol in Flammulina velutipes.The optimum chromogenic conditions were 2mL Folin-Ciocalteu reagent,2.4 mL 15%Na2CO3 at 45℃ for 70min.The method was accurate,stable and reliable for the determination of total polyphenol in Flammulina velutipes with greatly improved detection sensitivity.关键词
