补肾中药单体对氧化损伤内皮细胞中p66mRNA的调控作用
Regulating Effect of Tonifying Kidney Monomer on p66 mRNA of Oxidative Endothelial Cells

Abstract
Objective:To study the regulation of p66 mRNA of six kinds of tonifying kidney monomer:geniposidic acid,oleanolic acid,psoralen,sesamin,specnuezhenide,cinnamic acid on oxidative endothelial cells and to explore the mechanisms of its anti-oxidation and protection of vascular endothelial cells.Methods:EA.hy926 (human umbilical vein endothelial cells) were induced by H2O2 to be the oxidative stress damage model of vascular endothelial cells.After the intervention of different concentrations of geniposidic acid,oleanolic acid,psoralen,sesamin,specnuezhenide,cinnamic acid,p66 mRNA in the oxidative endothelial cells was detected by qPCR assay,Results:The superoxide dismutase (SOD) activity was decreased.The content of malondialdehyde (MDA) and reactive oxygen species was increased after the stimulation of H2O2.5 μM,50μM,500μM of geniposidic acid,psoralen,sesamin,cinnamic,especially acid specnuezhenide and oleanolic acid,inhibited the level of p66 mRNA expression level of oxidative vascular endothelial cells model.Conclusions:The effect of anti-oxidation and protection of vascular endothelial cells of tonifying kidney monomer is related with the regulation of p66 mRNA expression level.关键词
