Effects of Asparagus polysaccharides on growth, invasion and migration of human hepatocellular carcinoma cells under hypoxia

Abstract
Objective To observe the effect of Asparagus polysaccharides on the growth,invasion and migration of human hepatocellular carcinoma cells under hypoxia condition,and to explore the anti-tumor mechanism of Asparagus polysaccharides.Methods We cultured human hepatocellular carcinoma HepG2 cells,and added 500,1 000,1 500,2 000,3 000 and 4 000 μg/mL Asparagus polysaccharides to the 96-well culture plate.The control wells were added with the normal saline,then were put into the hypoxia incubator (95%N2,5%CO2,1%O2) for 24,48 and 72 hours.CCK-8 assay was used to detect the inhibition rate of Asparagus polysaccharides on human hepatocarcinoma cells.According to the Asparagus polysaccharides IC50,IC20,and IC10,we used the high-,medium-and low-concentrations of Asparagus polysaccharides to intervene HepG2 cells,which were then taken as the H-Asp,M-Asp,and L-Asp groups.The blank control group was not treated.After 24 h in hypoxia incubator,Transwell assay was used to detect the invasion and migration abilities of HepG2 cells.Results At different concentrations and different time,Asparagus polysaccharides had significant growth inhibition on HepG2 cells.The correlation analysis showed that the cell growth inhibition rate was positively correlated with the concentrations of Asparagus polysaccharides (r=0.980,P<0.05) with a dose-dependent manner.The number of invasive and migratory cells in the H-Asp,M-Asp,L-Asp groups was significantly less than that of the blank control group (all P<0.01).Conclusion Asparagus polysaccharides can significantly inhibit the growth of human hepatocellular carcinoma cells under hypoxia as well as effectively inhibit their invasion and migration.关键词
