Abstract
Objective To investigate the efficacy and safety of docetaxel and pemetrexed combined with platinum on patients with NSCLC after failure of gefitinib therapy.Methods One-hundred-and-ten patients with NSCLC after failure of gefitinib therapy were included and randomly (random number) divided into group A (n =55) and group B(n =55),which were treated with docetaxel combined with platinum or pemetrexed combined with platinum,respectively.The shortterm clinical effects,overall survival,progression free survival,EORTC QLQ-C30 score,KPS score and serious adverse effects were compared between the two groups.Results There was no significant difference in the objective response rate (ORR) or disease control rate (DCR) between the two groups(P ＞ 0.05).Group B showed longer progression free survival time than group A(P ＜ 0.05).No significant difference was found in overall survival time between the two groups (P ＞0.05).Nevertheless,both groups had higher EORTC QLQ-C30 scores and KPS scores after treatment (P ＜ 0.05),and group B had even higher scores than group A (P ＜ 0.05).The incidence of Ⅲ-v degree leucopenia,decreased hemoglobin,thrombocytopenia,nausea,vomiting,skin rash and liver function damage were all significantly lower in group B(P ＜0.05),yet no significant difference in the incidence of Ⅲ-Ⅳ degree diarrhea and hair loss were observed between the two groups(P ＞ 0.05).Conclusion The clinical effects of docetaxel and pemetrexed combined with platinum in treating NSCLC after failure of gefitinib therapy were comparable,but pemetrexed combined with platinum can effectively prolong progression free survival time,improve the quality of life and reduce the risk of serious adverse reactions.关键词
