关岭牛FoxO6基因原核表达载体的构建及生物信息学分析
Bioinformotion analysis of Guanling cattle FoxO6 gene and construction of its prokaryotic expression vector

Abstract
The experiment aimed to analysis functions of the FoxO6 gene.We obtained the CDS region of Guarding cattle FoxO6 gene by the method of PCR and then constructed the recombination plasmid pET32a-FoxO6 and analyzed the sequence of FoxO6 protein successfully.The CDS region of Guanling cattle FoxO6 gene had 942 bp,encodes 313 amino acids and the protein weights 33.60 kD.The protein was hydrophilic,the isoelectric point of FoxO6 protein was about 9.44 and had positive charges in vivo.The protein had mainly random coil and α-helix concerning the secondary structure prediction,no signal peptide and the transmembrane helical structure showing an intracellular protein,mainly existed in nucleus.The recombination expression protein of FoxO6 had His tag etc,encoded 492 amino acids and weights 53.0 ku.The experiment preliminary analyzed physic-chemical properties and structure characteristics providing the basis theoretical knowledge for studying function and mechanism of the FoxO6 protein in vivo .关键词
